Variations in levels of apolipoprotein E have been tied to the risk and progression of Alzheimer's disease . Our group has previously compared and contrasted the promoters of the mouse and human ApoE gene promoter sequences and found notable similarities and significant differences that suggest the importance of the APOE promoter's role in the human disease. We examine here three specific single-nucleotide polymorphisms within the human APOE promoter region, specifically at -491 , -427 , and at -219 upstream from the (...) +1 transcription start site. The -219 and -491 polymorphic variations have significant association with instance of AD, and -491AA has significant risk even when stratified for the APOEepsilon4 allele. We also show significant effects on reporter gene expression in neuronal cell cultures, and, notably, these effects are modified by species origin of the cells. The -491 and -219 polymorphisms may have an interactive effect in addition to any independent activity. DNA-protein interactions differ between each polymorphic state. We propose SP1 and GATA as candidates for regulatory control of the -491 and -219 polymorphic sites. This work's significance lies in drawing connection among APOE promoter polymorphisms' associations with AD to functional promoter activity differences and specific changes in DNA-protein interactions in cell culture-based assays. Taken together, these results suggest that APOE expression levels are a risk factor for AD irrespective of APOEepsilon4 allele status. (shrink)

BACKGROUND: Alzheimer's disease is intimately tied to amyloid-beta peptide. Extraneuronal brain plaques consisting primarily of Abeta aggregates are a hallmark of AD. Intraneuronal Abeta subunits are strongly implicated in disease progression. Protein sequence mutations of the Abeta precursor protein account for a small proportion of AD cases, suggesting that regulation of the associated gene may play a more important role in AD etiology. The APP promoter possesses a novel 30 nucleotide sequence, or "proximal regulatory element" , at -76/-47, from the (...) +1 transcription start site that confers cell type specificity. This PRE contains sequences that make it vulnerable to epigenetic modification and may present a viable target for drug studies. We examined PRE-nuclear protein interaction by gel electrophoretic mobility shift assay and PRE mutant EMSA. This was followed by functional studies of PRE mutant/reporter gene fusion clones. RESULTS: EMSA probed with the PRE showed DNA-protein interaction in multiple nuclear extracts and in human brain tissue nuclear extract in a tissue-type specific manner. We identified transcription factors that are likely to bind the PRE, using competition gel shift and gel supershift: Activator protein 2 , nm23 nucleoside diphosphate kinase/metastatic inhibitory protein , and specificity protein 1 . These sites crossed a known single nucleotide polymorphism . EMSA with PRE mutants and promoter/reporter clone transfection analysis further implicated PuF in cells and extracts. Functional assays of mutant/reporter clone transfections were evaluated by ELISA of reporter protein levels. EMSA and ELISA results correlated by meta-analysis. CONCLUSIONS: We propose that PuF may regulate the APP gene promoter and that AD risk may be increased by interference with PuF regulation at the PRE. PuF is targeted by calcium/calmodulin-dependent protein kinase II inhibitor 1, which also interacts with the integrins. These proteins are connected to vital cellular and neurological functions. In addition, the transcription factor PuF is a known inhibitor of metastasis and regulates cell growth during development. Given that APP is a known cell adhesion protein and ferroxidase, this suggests biochemical links among cell signaling, the cell cycle, iron metabolism in cancer, and AD in the context of overall aging. Recently, we have seen the emergence of the international laboratory in scientific research. These laboratories, characterized by internationally distributed members working to accomplish a unified goal, provide advantages such as cost savings and access to facilities and equipment. However, maintaining responsible conduct of research (RCR) in an international laboratory is complicated by the requirement for technology-mediated communication, lack of trust between local and distant group members, and cultural heterogeneity among lab members. Here we discuss issues we experienced while working in (...) a international laboratory as part of the Singapore-M.I.T. Alliance for Research and Technology, and from our experience, we propose guidelines to maintain RCR in an international laboratory. With proper oversight and cognizance of RCR, work in an international laboratory harbors great benefit for scientists and provides unique opportunity to exchange ideas, data, and insight with scientists of distinct cultural backgrounds. Major hallmarks of Alzheimer's disease include brain deposition of the amyloid-beta peptide , which is proteolytically cleaved from a large Abeta precursor protein by beta and gamma- secretases. A transmembrane aspartyl protease, beta-APP cleaving enzyme , has been recognized as the beta-secretase. We review the structure and function of the BACE1 protein, and of 4129 bp of the 5'-flanking region sequence of the BACE1 gene and its interaction with various transcription factors involved in cell signaling. The promoter region and 5'-untranslated (...) region contain multiple transcription factor binding sites, such as AP-1, CREB and MEF2. A 91 bp fragment is the shortest region with significant reporter gene activity and constitutes the minimal promoter element for BACE1. The BACE1 promoter contains six unique functional domains and three structural domains of increasing sequence complexity as the "ATG" start codon is approached. Notably, the BACE1 gene promoter contains basal regulatory elements, inducible features and sites for regulation by various important transcription factors. Herein, we also discuss and speculate how the interaction of these transcription factors with the BACE1 promoter can modulate synaptic plasticity, neuronal apoptosis and oxidative stress, which are pertinent to the pathogenesis and progression of AD. Nurses have moral obligations incurred by membership in the profession to participate knowingly in health policy advocacy. Many barriers have historically hindered nurses from realizing their potential to advance health policy. The contemporary political context sets additional challenges to policy work due to polarization and conflict. Nursing education can help nurses recognize their role in advancing health through political advocacy in a manner that is consistent with disciplinary knowledge and ethical responsibilities. In this paper, the authors describe an exemplar of (...) Elizabeth Barrett's “Power as Knowing Participation in Change” theory as a disciplinary lens within a doctoral nursing health policy course. Barrett (radically) emphasizes “power as freedom” instead of “power as control.” This approach is congruent with nursing disciplinary values and enhances awareness of personal freedom and building collaborative relationships in the policy process. The theory was used in concert with other traditional policy content and frameworks from nursing and other disciplines. We discuss the role of nursing ethics viewed as professional responsibility for policy action, an overview of Barrett's theory, and the design of the course. Four student reflections on how the course influenced their thinking about policy advocacy are included. While not specific to policymaking, Barrett's theory provides a disciplinary grounding to increase students' awareness of freedom and choices in political advocacy participation. Our experience suggests that Barrett's work can be fruitful for enhancing nurses' awareness of choices to participate in change across settings. (shrink)

They are not, first of all, phenomenologists or existentialists in the strict sense, by which I mean they do not hold the sceptical epistemological premises usually associated with these labels. Existentialists deny the classical doctrine of truth according to which truth is the adaequatio ad rern of the intellect. Instead they maintain that when I look at, say, an egg, and say to myself, " Here I am, one being among others like this egg here ", I am being naive because I have not yet seen that the distinction between myself and the egg arises within t:he phenomenal field and therefore cannot be uncritically assumed in an account of knowing. The subject-object distinction, they say, is not one more distinction between objects, like that between the egg and the chicken it came from; it cannot be understood as an intellectual grasp of one individual substance by another individual substance. For this reason, according to the existentialists, we must not think of knowledge as an overcoming of the isolation of self-consciousness, but rather as the origination of a distinction, a negative act identical with that of selfconsciousness in its self-transcendence. Awareness of the egg is not a grasp of the egg in itself so much as a self-conscious mitosis which incidentally constitutes the egg as object.4 The realist answer, as Chesterton said, is that eggs are eggs. I let it sit around for many months, fearing, as did the author, to "[row] out over the great ocean (...) of material" (pp. xix-xx), but once I had set sail I found myself in enjoyable, if sometimes irascible, company and I was unwilling to jump ship. Girardot is in full command of his ark of fascinating and diverse creatures, and his main argument is never lost in the details. His writing is lucid and the text is broken up into manageable blocks by subheadings with intriguing titles, rendering each segment ehipassiko, encouraging us to keep reading.A book of this importance and weight (about 1.5 kg) deserves a review article more than just a review, or, better, a discussion panel at a major academic conference. This journal is notorious for reviews of astonishing length, but I would like my review to be short enough that it will actually be read, so that it will entice you to read the book. I will not attempt a summary nor will I critically address all the issues that are brought up. Instead, I will try to show why I regard this book as an instant classic and even, I would say, a watershed in the meta-science of sinological and Religionswissenschaftlich self-reflection.We all know James Legge as the perpetrator of The Chinese Classics, the multivolume opus that, in the 1960 Hong Kong University Press revision, dominates the Chinese religion(s) section of our personal libraries. Since there is nothing like it, we consult it, but we pretend we do not, for, after all, it's out of date, surely, and Legge was an old fuddy-duddy—Scottish, a Protestant missionary, and, worst of all, a Victorian. Girardot seems to have had somewhat this opinion of Legge when he assayed a brief biography of him, but he discovered that his "Legge work" (as he calls it) opened the door to a pivotal era in academe and world culture. Once the door to the long-neglected cupboard had been opened, all kinds of fascinating objects came tumbling out. "Oh, so that's how it used to be, and that's why it's like this today!" No scholar of Chinese religion(s) can afford to ignore Girardot's extensive and insightful ruminations on the state of the field then and now.The point of the book is summed up on page 531: "[Legge] is an outstanding [End Page 276] example of the difficult nineteenth-century passage from religious discourse to what Edward Said calls the 'secular criticism' of the modern world." To study Legge is to study the origins of our discipline. He may have been a fuddy-duddy; he was certainly a workaholic, putting his Sassenach colleagues at Oxford to shame by sleeping little and rising in the middle of the night to persist in his hermeneutic labors. But he was also a student of the heathen land he went to convert, and he tried his best not only to see (the presumed universal) God in the ancient texts but also to find something noble in apparent atheists and agnostics such as Confucius. In his interaction with Max Mueller (and this book is as much about Mueller as it is about Legge) he both agreed and disagreed, in a complex choreography, with the comparative method underpinning The Sacred Books of the East. He was a Scotsman's Scotsman, a stag at bay, dourly standing his ground (as a Congregationalist, and therefore a nonconformist both north and south of the border) against, first, the London Missionary Society and, later, the prevailing academic and ecclesiastical climate at Oxford.No armchair orientalist, as were some of his colleagues across the Channel, he befriended native informants and studied the indigenous commentaries (mostly Qing) to which they referred him, so that his translations, although not as fluid as Waley... Background Biobank participants often do not understand much of the information they are provided as part of the informed consent process, despite numerous attempts at simplifying consent forms and improving their readability. We report the first assessment of biobank enrollees’ comprehension under an "integrated consent” process, where patients were asked to enroll in a research biobank as part of their normal healthcare experience. A number of healthcare systems have implemented similar integrated consent processes for biobanking, but it is unknown how (...) much patients understand after enrolling under these conditions. Methods: We recruited patients who enrolled in a biobank while in a healthcare setting when receiving ordinary care. We assessed knowledge of consent materials using 11 true/false questions drawn from a well-known biobank knowledge test. After reviewing the results from 114 participants, we revised the consent form and repeated the knowledge assessment with 144 different participants. Results: Participants scored poorly on the knowledge test in both rounds, with no significant differences in overall scores or individual items between the rounds. In Phase 1, participants answered 53% of the questions correctly, 25% incorrectly, and 22% “I don’t know.” In Phase 2, participants answered 53% of questions correctly, 24% incorrectly, and 23% “I don’t know.” Participants scored particularly poorly on questions about data sharing and accessing medical records. Conclusions: Enrollees under an integrated consent model had significant misunderstandings that persisted despite an attempt to improve information specifically about those topics in a consent form. These results raise challenges for current approaches that attribute misunderstanding to overly complex consent forms. They also suggest that the pressures of the clinic may compound other problems with patient understanding of biobank consent. As health systems increasingly blend research and care, they may need to rethink their approach to educating patients about participation in a biobank. (shrink)

Sometimes ordinary disagreements become deep as a result of epistemic injustice. The paper explores a hitherto unnoticed connection between two phenomena that have received ample attention in recent social epistemology: deep disagreement and epistemic injustice. When epistemic injustice comes into play in a regular disagreement, this can lead to higher-order disagreement about what counts as evidence concerning the original disagreement, which deepens the disagreement. After considering a common definition of deep disagreement, it is proposed that the depth of disagreements is (...) best understood as a matter of degree. Then, a case study of real-life disagreement is introduced: the disagreement about whether racism is a significant issue in the Netherlands, illustrated by the tradition of ‘Black Pete’. It is argued that there is disagreement about what counts as evidence in the case study because of two forms of epistemic injustice: testimonial and hermeneutical injustice. Specifically, there is disagreement about epistemic principles concerning whether private first-personal experience of racism is a weighty source of evidence in this domain, whether victims of racism count as important testifiers in this domain, and how to assess testimony that is not intelligible to you because it employs concepts and terminology you are unfamiliar with. Pp. xii + 263.Roger-Pol Droit's recently translated study, The Cult of Nothingness: The Philosophers and the Buddha, is not a book about Buddhism per se. Rather, it is a rich and theoretically (...) sophisticated overview of the reception of Buddhism in Europe that deserves shelf space alongside Wilhelm Halbfass' India and Europe and Raymond Schwab's The Oriental Renaissance. But while some of the figures Droit covers may be familiar from these other studies in the history of orientalism, Droit's book has its own unique narrative thread. Specifically, Droit wishes to examine the once widespread notion that Buddhism is a nihilistic religion, a religion that worships nothingness. His central thesis is that this conception of a nihilistic religion really wasn't about Buddhism at all, but "that Europe... was speaking only of herself." Droit sees in the cult of nothingness the subconscious expression of a crisis of foundations among nineteenth-century European intellectuals, as well as a "hidden laboratory for the theoretical development of European nihilism."Along the way to illustrating this thesis, Droit leads a masterful and extremely entertaining tour of the opinions of early Buddhologists and Eastward-looking philosophers, some of whom have seldom been documented elsewhere. He also provides an enormous bibliography of European writings on Buddhism from 1638 to 1890. In the eighteenth and early nineteenth centuries, the Buddha was often identified as one of many gods belonging to an inchoate and primitive world of mythical concordances. Thus, George Stanley Farber wrote in 1816 that "the primeval Buddha... is the same as Vishnu, or Shiva, or Osiris." Writers in this genre were especially fond of identifying the Buddha with the god Mercury. The sole reason, it seems, was simple linguistic confusion-the word for the planet Mercury in Sanskrit is budha, one letter short of buddha. Furthermore, the preeminent Indologist Sir William Jones posited the identity of the Norse god Woden and Buddha; this was later taken up by A. W. Schlegel in his Indische Bibliothek, and eventually influenced the publication of such unforgettable titles as Holmboe's Traces du Buddhisme en Norvège avant l'introduction du Christianisme. Jones elsewhere hinted that the Buddha might have been of Ethiopian, rather than Indian origin, and this set off another wave of speculation. To reconcile theories of a primordial god named Buddha with new evidence that the Buddha had been a man who had lived at a relatively recent date, for a short time a theory of two Buddhas emerged, first suggested by Antione Augustin Giorgi, and later taken up by Jones himself.Droit sees Hegel as the single figure most responsible for establishing the link between Buddhism and nothingness that endured in the nineteenth-century philosophical imagination. Droit painstakingly charts Hegel's changing attitudes toward Buddhism over the course of his lectures from 1822 to 1831. In spite of, or perhaps because of, Hegel's familiarity with all of the most recent secondary works on Buddhism, he had great difficulty in integrating Buddhism into his philosophical grand [End Page 577] narrative. But his most enduring interpretation of Buddhism came in 1827, when he asserted that for Buddhism, "Saintliness consists in man uniting himself with God, with nothingness, with the absolute in this destruction, in this silence.... Man must make himself nothing." Hegel did not get this idea from his contemporaries, but from an earlier stratum of writing on Buddhism, that of seventeenth- and eighteenth-century Jesuit missionaries. After this portrayal of Buddhism became prevalent in European philosophical circles, an extraordinary debate developed. The notion of a religion that worships nothingness was so contrary to common sense that it threatened the Enlightenment conviction that there existed a single, universal human nature. Jules Barthélmy Saint-Hilaire summed it up quite clearly: "In the presence of a phenomenon so curious and so deplorable... one might ask... if, in those climates where life is so horrific and nothingness is worshiped instead of God, human nature is... This is the first of three volumes which will contain all of Locke's extant philosophical writings relating to An Essay Concerning Human Understanding, not included in other Clarendon editions like the Correspondence. It contains the earliest known drafts of the Essay, Drafts A and B, both written in 1671, and provides for the first time an accurate version of Locke's text. Virtually all his changes are recorded in footnotes on each page. Peter Nidditch, whose highly acclaimed edition of An Essay (...) Concerning Human Understanding was published in this series in 1975, used pioneering editorial techniques in his compilation of Volume 1. Most of the work was completed before his tragically early death in 1983. Volumes 2 and 3, almost wholly the work of G. A. J. Rogers will contain the third extant draft of the Essay, the Epitome and the Conduct of the Understanding. They will also include a History of the Writing of the Essay, together with other shorter writings by Locke. (shrink)

Political philosophy was once dominated by discussion of the virtues of character and their importance to the good life and the good society. Contemporary political philosophers, however, following the towering influence of John Rawls, have primarily focused on a single virtue of institutions: justice, while largely avoiding controversial claims about the good life. As a result, political philosophy lacks a unified account of the virtues of institutions and the virtues of character. More importantly, we lack an understanding of the connection (...) between the just society and the good life. This book begins to mend this broken seam, which, on reflection, lies at the heart of our deepest political problems. The book’s central argument is that the virtues of character require institutions, while good institutions enable persons to live together virtuously. Political institutions are a necessary precondition for the moral agency exhibited by the virtues. But good institutions enable virtue to grow, while helping to constrain the excesses of vice. On this view, justice emerges primarily as a virtue of character, not institutions. The just person balances conformity to existing institutional norms with practically wise reforms. Ultimately, this book concludes that the just society and the good life are linked through the common good of virtuous activity within the shared institutions of a political community. (shrink)

In this prcis we focus on phenomena central to the reaction against similarity-based theories that arose in the 1980s and that subsequently motivated the approach to semantic knowledge. Specifically, we consider (1) how concepts differentiate in early development, (2) why some groupings of items seem to form or coherent categories while others do not, (3) why different properties seem central or important to different concepts, (4) why children and adults sometimes attest to beliefs that seem to contradict their direct experience, (...) (5) how concepts reorganize between the ages of 4 and 10, and (6) the relationship between causal knowledge and semantic knowledge. The explanations our theory offers for these phenomena are illustrated with reference to a simple feed-forward connectionist model. The relationships between this simple model, the broader theory, and more general issues in cognitive science are discussed. (shrink)
